I have downloaded a .uif file, and used uif2iso from OS4Depot to decode/convert? it to an .iso file. When looking at the .iso file, I get a read error, so I looked for an alternative.

I noticed that ISO-o-Matic can handle .uif files, but when I try to use it I get the error message 'object is too large'
I have set my stack to 132768, and then 5132768, and both give me the same error. The .uif file size is 130,970,297, and I have 334,528,512 freemem on workbench.

Anyone any idea what I am doing wrong?

ISO-o-Matic will never return an ERROR_OBJECT_TOO_LARGE error itself. Likely this error is returned by the filesystem to ISO-o-Matic. If you kept the partial ISO file generated you could check if it is close to either 2GB or 4GB. The file size limit for SFS\0 is just under 4GB and it will return this error if a program tries to write more data to a file.

Since UIF is a compressed file format the generated ISO file will likely be much larger than the original file.

The .uif file is 130,970,297 in size and the .iso file (with the read errors) is 138,280,960. Now I am not a programmer, so I am not up on what those file sizes are in Mb but, I don't think it is anywhere near 2Gb.
The .uif file only contains pdf files so there is probably not much compression anyway.
