Haven't tried it. I was under the impression that was for communication with a real Amiga.

In my old AF setups I always used the "AmigaExplorer" to access the Amiga side from the PC side when using AmigaForever.
I would assume that it still works in AF 2016, but have not tried it yet.
I have to finish the the Amiga-OS installation first, as time permits...
